Reacts with a PI-linked glycoprotein, Mw 18-20 kDa found on all types of leucocytes including platelets. The antigen is expressed on many haemapoietic and non-haemapoietic cells. MEM-43 has been shown to stimulate NK activity. CD59 is expressed by erythrocytes and therefore staining in whole blood systems requires a high concentration of antibody. The epitope recognised by MEM-43 is lost after reduction, therefore, non-reducing conditions are required for western blotting techniques (2, 5).

ELISA. Immunoprecipitation. Western blot (non-reducing conditions). Flow Cytometry (Use 10 μL of 1/5-1/10 diluted antibody to label 106 cells in 100 μL). Immunohistochemistry on Frozen and Paraffin Sections: Requires antigen retrieval using heat treatment methods prior to staining of paraffin sections, Sodium citrate buffer pH 6.0 is recommended for this purpose. Positive control: Tonsil. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

CD59, an LY6 like protein expressed in human lymphoid cells, regulates the action of the complement membrane attack complex on homologous cells. It is a potent inhibitor of the complement membrane attack complex action. It acts by binding to the C8 and/or C9 complements of the membrane attack complex, thereby preventing incorporation of the multiple copies of C9 required for complete formation of the osmolytic pore. This inhibitor appears to be species-specific.
