RNF34, also known as RING finger protein 34, RING finger protein RIFF, FYVE-RING finger protein Momo, Human RING finger homologous to inhibitor of apoptosis protein, hRFI, Caspases-8 and -10-associated RING finger protein 1, CARP-1 and Caspase regulator CARP1, is a novel modulator of NF-kappaB activation. RNF34 possesses E3 ubiquitin protein ligase activity and has been shown to regulate the levels of CASP8 and CASP10 by targeting them for proteasomal degradation. RNF34 protects cells against apoptosis induced by TNF and may bind phosphatidylinositol phosphates. RNF34 is localized to both the nucleus and the cytoplasm. In the nucleus, a distinct speckled pattern is observed by immunofluorescence microscopy. In the cytoplasm, the protein is associated with membranes. RNF34 is expressed ubiquitously and can be detected in heart, brain, liver, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, ovary, colon and leukocytes.
