In humans, attachment of ubiquitin to Histone protein 2A (H2A) on lysine residue 119 (H2AK119Ub) is one of the most prevalent histone marks, and is estimated to occur on 5-15 % of histones genome-wide4. In general, H2AK119Ub is characteristic of repressed developmental genes, and is also tightly linked to levels of other histone marks such as lysine methylation and acetylation. H2AK119Ub is attached to histones by the Really Interesting New Gene (RING) ubiquitin E3-ligase Polycomb Repressive Complex 1 (PRC1), and removed by the Polycomb Repressive-Deubiquitinase (PR-DUB) complex, which plays a vital role in regulating H2AK119Ub histone marks, and mutations in human PR-DUB genes lead to a spectrum of malignancies. Histone H2AK119ub antibody (pAb) was raised in a Rabbit host. It has been validated for use in Dot blot and Western blot, it has been shown to react with Human samples, but it is predicted that it will react with a wide range of sample types.
