This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for sCD30L has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sCD30L present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for sCD30L is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of sCD30L bound in the initial step. The color development is stopped and the intensity of the color is measured.

The human CD30 ligand, CD153, is a 40kDa Type II transmembrane glycoprotein. It interacts with its receptor, CD30, as a homotrimer Soluble trimeric CD153 binds to membrane-anchored CD30 with a relatively high affinity and is very effective in triggering cell death and TNFα. CD153 is a member of the TNF superfamily which is primarily expressed by activated T cells, B cells and monocytes. Its function in programmed cell death has been proven, as well as its involvement in disorders like a variety of lymphoid malignancies like Hodgkin’s disease and anaplastic large cell lymphoma.
