    Highly purified Cathepsin G is bound to microwells . Antibodies to this antigen, if present in diluted serum, bind in the microwells. Washing of the microwells removes unbound serum antibodies. Horseradish peroxidase (HRP) conjugated antihuman IgG immunologically bind to the bound patient antibodies forming a conjugate/antibody/antigen complex. Washing of the microwells removes unbound conjugate. An enzyme substrate in the presence of bound conjugate hydrolyzes to form a blue color. The addition of an acid stops the reaction forming a yellow endproduct. The intensity of this yellow color is measured photometrically at 450 nm. The amount of colour is directly proportional to the concentration of IgG antibodies present in the original sample.

    Hintergrund
    The cathepsins belong to a group of intracellular proteases mainly found in lysosomes, especially of the spleen, the liver and the kidney. Cathepsin G is a serine protease and a further pANCA antigen. It participates in a great part in the destruction of osteid tissue as a result of its hydrolytic properties. The autoantibodies against Cathepsin G occur mainly in collagenosis and other related inflammatory rheumatic diseases, e.g. SLE, Sjögren syndrome and Felty syndrome.
