Bioassay: Activity Range Tested by MTT/Actinomycin D Cytolysis Assay using L929 indicator cells
Specific Activity: 0.25 - 5.0 x 10^9 Units /mg
ED50: 2 - 40 pg/ml
Observed Dose response: 250 fold
The Unit is defined as the amount of material required to stimulate a half-maximal response at cytokine saturation. Since applications vary, titrate the reagent to obtain optimal results. - Produktmerkmale
- Recombinant rat TNF is supplied as a frozen liquid comprised of 0.22 µm sterile-filtered aqueous buffered solution containing 2 mg/ml biotechnology grade, low endotoxin bovine serum albumin, with no preservatives. Recombinant rat TNF was purified by immunoaffinity chromatography and yielded a TNF protein fraction which was found to be > 95% pure by SDS-PAGE analysis, and an absorbance assay based on the Beers-Lambert law.

ELISA Standard: Recombinant rat TNF is useful as a quantitative standard for measuring rat TNF protein levels in a TNF specific sandwich ELISA with purified TN3-19.12 antibody (Cat. No. 557516) as a capture antibody and biotinylated rabbit anti-mouse/rat TNF antibody (Cat. No. 557432) as the detection antibody. To obtain linear standard curves, doubling dilutions of rat TNF standard from ~10 ng - 0.1 ng/ml should be included in each ELISA plate.
Note: This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. For measuring rat TNF in serum or plasma the rat TNF OptEIA™ Set (Cat. No. 558535) and rat TNF OptEIA™ Kit (Cat. No. 560479) are specially formulated and recommended. For specific methodology, please visit our web site, www.bdbiosciences.com, and go to the protocols section or the chapter on ELISA in the Immune Function Handbook. - Kommentare
