- MEK1 double mutant human is a recombinant protein containing a polyhistidine tag expressed in Sf9. This double mutant protein contain alanine substitutions at S218A and S222A rendering it inactive. Analysis by SDS-PAGE resulted in a pattern consistent with purified MEK1 and was estimated to be greater than 90% pure.

Application Note: Human MEK1 mutant recombinant protein has been tested in SDS-Page and is suitable as a negative control protein for immunoassays using antibodies targeting the critical MEK1 phosphorylated S218 or S222 residues. For western blot use at 50 ng or less. For other assays concentration is user optimized.

- Background: MEK1 isoform double mutant recombinant protein is modified to put alanine residues at two key activation sites, Serine 218 and Serine 222. Mitogen-activated protein kinase kinase 1, also known as MKK or MEK, is an integral component of the MAP kinase cascade that regulates cell growth and differentiation. This pathway also plays a key role in synaptic plasticity in the brain. Activated MEK 1 acts as a dual specificity kinase phosphorylating both a threonine and a tyrosine residue on MAP kinase. The inactive double mutant MEK1 recombinant protein is ideal for investigators involved in Neuroscience, Cell Signaling and Cancer Research.
